Using the bioWORLD’s α-amylase Purification and Assay kit the enzyme can be purified from any bacteria/fungal extract or exudate and purified to homogeneity by ammonium sulphate precipitation followed by affinity chromatography. The α-amylase adsorption to starch immobilized Separopore® 4B-CL (Agarose) allows α-amylase to bind to starch more tenaciously, and allows instant elution using 2% (w/v) dextrin. Electrophoresis analysis can confirm the purity of the enzyme.
For extracellular enzyme, the exudate can be precipitated using 60% Ammonium Sulfate, dialyzed and applied to the Affinity Matrix.
The Separopore®4B – Amylase Affinity Matrix is supplied in 10% ethanol. Wash the gel after packing in a column with 1x Phosphate Buffer (Extraction Buffer).
