AMPD (2-Amino-2-methyl-1,3-propanediol) buffer has a concentration of 0.2M and a pH value of 9.5. It has a boiling point 151°C, a melting point range of 100 - 110°C, and a density of 1.126 g/cm3. This buffer is used as a reaction buffer for enzymes with an activity in the basic pH range and in isotachophoresis (ITP) of proteins. ITP is a form of electrophoresis and is a technique in analytical chemistry used for selective separation and concentration of ionic analytes. AMPD is a component in SDS-gradient gel electrophoresis system that separates polypeptides in the molecular weight range of 1,500 – 100,000. The pH can be adjusted with NaOH or HCl, with no crossing of the desired pH to then backtrack since this may result in altering the salt concentration.
