Background: IGFBP-1 is one of the six homologous proteins that specifically bind insulin-like growth factors and modulate their mitogenic and metabolic actions. The function of this protein is not completely defined. However, several studies demonstrate that it inhibits IGF binding to cell surface receptors and thereby inhibits IGF-mediated mitogenic and cell metabolic actionsInsulin Like Growth Factor Binding Protein modulate IGF activities by increasing their plasma half lives and by inhibiting or promoting the interactions of IGF with receptors on certain target cells. In addition these binding proteins provide a reservoir for IGF in pericellular spaces. Some IGF-BPs also have stimulating effects in vitro and some may inhibit the growth of cells

Description: Recombinant Human Insulin Like Growth Factor Binding Protein-1 is a single glycosylated polypeptide chain containing 218 amino acids and having a molecular mass of 28806 Dalton. The IGFBP-1 is purified by proprietary chromatographic techniques.

Physical Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation: IGFBP-1 was lyophilized with 10mM HCL.

Solubility: It is recommended to reconstitute the lyophilized IGFBP-1 in sterile 18MO-cm H2O not less than 100 g/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized IGFBP-1 although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuIGFBP-1 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Purity: Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Anion-exchange FPLC. (c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Biological Activity: IGFBP-1 is fully biologically active when compared to standards. Inhibition of IGF-I stimulated proliferation of NUC-1 cells: IC50 ~ 100 ng/ml Inhibition of serum-induced stimulation of DNA synthesis in chick embryo fibroblast primary cultures: IC50 (1% serum) ~300 ng/ml.

Endotoxin: Less than 0.1 ng/ g (IEU/ g) of IGFBP-1 .

Protein content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm. 2. Analysis by RP-HPLC, using a standard solution of IGFBP-1 as a Reference Standard.
