Background: The SS-A/Ro particle contains a hY RNA (human cytoplasmic RNA) and associated proteins: a 60 kDa and a 52 kDa protein. The 52 kDa protein is not directly bound to the hY RNA but to the 60 kDa protein. It seems that it is sometimes associated, sometimes not. The Ro antigen is found in the cytoplasm but also in the nucleus. The role of the SS-A/Ro particle in the cell is not known yet. An autoantibody against SS-A/Ro52 (Ro52) is most frequently found in the sera of patients with Sjogren\\\'s syndrome, systemic lupus erythematosus, and congenital heart block from anti-Ro52 Ab-positive mother. However, the physiological function of the autoantigen SS-A/Ro52 has not yet been elucidated. Ro52 protein plays a role in T cell activation. Overexpression of SS-A/Ro52 in Jurkat T cell resulted in enhanced IL-2 production following CD28 stimulation.

Description: Recombinant Human SS-A/RO-52 produced in E.Coli is a single, non-glycosylated, polypeptide chain having a molecular mass of 52kDa. Recombinant SS-A/RO-52 is purified by proprietary chromatographic techniques.

Physical Appearance: Sterile Filtered clear solution.

Formulation: RO-52/SS-A , at a concentration of 1mg/ml, contains 6M Urea, 500mM NaCl and 10mM Tris pH8.0.

Stability: RO-52/SS-A although stable at 4oC for 3 weeks, should be stored desiccated below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Purity: Greater than 90.0% as determined by: (a) Analysis by RP-HPLC. (b) Anion-exchange FPLC. (c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Endotoxin: Less than 0.1 ng/ g (IEU/ g) of RO-52/SS-A.

Protein content: Protein quantitation was carried out by using a 0.25 - 2.0 mg/ml Bradford assay vs. BSA
