Background: Human UBC9 Ubiquitin-like protein SUMO-1 conjugating enzyme is also called UBE2I. rHuUBC9 Mediates the conjugation of the ubiquitin-like protein SUMO-1 to a variety of proteins including RanGAP1, IkappaBalpha, and PML without the requirement of an ubiquitin ligase (E3).

Description: Human Ubquitin Conjugating Enzyme 9 (Ubc9) is a member of the E2 family and is specific for the conjugation of SUMO to a variety of target proteins. SUMO conjugation to target proteins is mediated by a different, but analogous, pathway to ubiquitinylation. This E2 is unusual in that it interacts directly with protein substrates that are modified by sumolyation, and may play a role in substrate recognition. Ubc9 can mediate the conjugation of SUMO-1 to a variety of proteins including RanGAP1, I?Ba, and PML without the requirement of an E3 ligase Recombinant Human Ubiquitin Conjugating Enzyme 9 produced in E.coli is a 19.5 kDa protein containing 171 amino acids. Recombinant Ubc9 protein contains 6xHis tag and is purified by proprietary chromatographic techniques.

Physical Appearance: Sterile Filtered whilte lyophilized powder.

Formulation: Lyophilized from a 0.2 m filtered concentrated (1mg/ml) solution in 1X PBS and 1mM DTT, pH 7.5.

Solubility: It is recommended to reconstitute the lyophilized Human Ubc9 in sterile water not less than 100 g/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized Recombinant Ubc9 although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rhUbc9 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Purity: Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by SDS-PAGE.
