Recombinant Mouse Interleukin-13 (IL-13)

Background: Interleukin-13 (IL-13) is a multifunctional cytokine whose net principle action is to diminish inflammatory responses. Interleukin (IL)-13 is an important cytokine secreted from type 2 helper T lymphocytes. It is essential for modulating IgE synthesis by human B cells. Previous studies showed that polymorphisms in the IL-13 gene were associated with serum total IgE or allergic asthma. Interleukin (IL)-13 is a pleiotropic cytokine, which shares many biological functions with IL-4. The receptor subunits of IL-13 consist of IL-4Ralpha, IL-13Ralpha1 and IL-13Ralpha2.

Description: Recombinant interleukin-13 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 110 amino acids and having a molecular mass of 12127 Dalton. Mouse IL-13 is purified by proprietary chromatographic techniques.

Physical Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation: The protein (1mg/ml) was lyophilized with no additives.

Solubility: It is recommended to reconstitute the lyophilized Recombinant IL-13 in sterile 18MO-cm H2O not less than 100 g/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized IL-13 although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution IL-13 should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Purity: Greater than 98.0% as determined by: (a) Analysis by RP-HPLC. (b) Anion-exchange FPLC. (c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and found to be Met-Pro-Val-Pro-Arg.

Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Biological Activity: ProSpec\\\'s Recombinant IL-13 is fully biologically active when compared to standard. The ED50 was determined by the dose dependent prolifiration of TF-1 cells and was found to be < 4ng/ml, corresponding to a specific activity of >2.5 x 105.

Endotoxin: Less than 0.1 ng/ g (IEU/ g) of recombinant mouse IL-13.

Protein content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm using the absorbency value of 0.69 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a calibrated solution of Interleukin-13 as a Reference Standard.
